How to set up & program these communication module to communicate with serial devive through non-protocol ascii code.

I believe they can only master or slave DirectNET or Modbus, and can slave as K-Sequence
To do what you want, I believe your only options are
1. to use the local CPU Port 2 on a 250 (master only) or 260 (master or slave) configured as non-sequence protocol or else
2. use a Basic Co-Processor module
